Levitra (vardenafil) for treatment of ED is rapidly absorbed after oral administration. The drug has bioavailability of about 15%. The max concentration in plasma is reached between half an hour and 2 hours after and oral dose.
The drug is metabolized by CYP3A4 (Cytochrome P450 3A4 one of the most important enzymes involved in the metabolism) and the M1 metabolite has 7% of the total activity.
The drug is eliminated after 4-6 hours, mostly as metabolites in feces (91-95%) and in urine (2-6%).
It is proved by studies that hepatic function impairment when taking this drug is connected with increased plasma levels. So the starting dose for such patients with moderate hepatic impairment should be 5mg, it is also recommended not to exceed the dose more than 10mg.
For men over 65 years the concentration of the drug in plasma and AUC (Area under the Curve) values which determine other pharmacokinetic parameters, such as clearance or bioavailability are increased 34% and 52% in comparison with men 18 to 45 years old.
